May grunwald giemsa stain is Romanowsky stain.They are composed of two components azure B and Eosin Y. Azure B bind with anionic parts of cell-like DNA and stain them blue. While Eosin Y binds with cationic parts of cell-like protein, it stains them the red color.

Giemsa stain was first discovered by Gustav Giemsa in 1902 It is composed of polychromed methylene blue and eosin. These components allowed simultaneous differential staining of cells.

What is the optimun PH of may grunwald giemsa stain solution ?

Optimum PH is required for Giemsa stain. An Alkaline PH accentuates the basic dye staining. Therefore Optimum PH is 6.8 for staining all components of cells.

  1. May Grunwald Stain powder
  2. Giemsa Stain powder
  3. Conical Flask
  4. Methanol (acetone free)

How to prepare giemsa stain working solution (giemsa stain composition)

  1. Add the weighed amount of Grunwald stain power in the conical flask. Add 100 methanol and mix well.
  2. Add the weighed amount of Giemsa stain is another conical flask and add 100 ml methanol and dissolve it.
  3. Warm both flask at 50C temperature for 15 minutes. Note: Mix well during warming.
  4. Both stains filter in clean bottles and store at room temperature in the darkroom.

May grunwald giemsa stain procedure (mgg stain)

  1. Prepare the smear and dry it.
  2. Fix the smear in absolute methanol for 4 to 5 minutes, after this dry it.
  3. Place the Giemsa stain 1:10 buffer on slid for 1 hour.
  4. Wash with water, blot, and dry.
  5. Examine under oil emersion lense.

How to dilute may grunwald giemsa

Giemsa stain diluted with 1:10 of water. This means 1 part of the stain was added in 10 parts of water.
