A platelet count blood test is important for the calculation of platelet in the blood. Like other blood tests Hb, CBC, LFT (liver function test), RFT (Renal function test), and platelet can be counted by two methods.

  • Manual Method
  • Automated method

Note: Both methods are used but the manual or visual method is more reliable than an electronic particle counter.

Requirements for Platelet count blood test

The following five are the requirements for platelet count blood tests.

  1. Platelet Diluting Fluid: 1% ammonium oxalate is used plt diluting fluid. It is 1% ammonium, oxalate is prepared by adding 1 gram of ammonium oxalate with 100 ml water.
  2. Improved Neubauer chamber:
  3. WBC pipette
  4. Microscope
  5. Petri Dish with moist cotton or tissue paper.

how do you perform a total platelet count manually?

Here’s the steps to perform total platelet count manual.

  1. First, prepare the 1:20 of blood with dilution.
  2. Place the coverslip on the improved Neubauer chamber, then charge it.
  3. Now cover the chamber in a Petri dish (To avoid the drying of the sample) for 15 to 20 minutes.
  4. After 20 minutes, now place the Neubauer chamber on the stage of the microscope and read at objective 40 x.
  5. Count the small high refractile discs in the central large square area of the Neubauer chamber.

Note: At least 200 platelets should be counted.

Manual Platelet count formula neubauer chamber

  • Central large area: 1 mm2
  • The volume of 1 large square area: 0.1 mm
  • Dilution: 1 in 20
  • Number of platelets: N

Platelet counting Formula: N* 20 * 10 9

Platelet count normal value

The platelet normal range for males and females is 150 to 340 * 109 .

what causes a high platelet count?

A high level of platelet is called thrombocytosis. It occurs in infection, hemolytic anemia, and iron deficiency.

What are the causes of decreasing platelet in the blood?

Normally platelet is normal in range. The lower level of the platelet is known as thrombocytopenia. but their range decrease in some diseases like Leukemia, Anemia, Bone marrow disease, and viral infection like Hepatitis A virus, Hepatitis B virus, and Dengue fever.

What is the KEY function of platelets in the body?

Platelets are colorless small fragments of megakaryocytes. Their main function is to stop the bleeding from the body, as they produce clots on the cut site.

How to increase platelet count in dengue?

1. Avoid alcohol
2. Exercise
3. Eat meat
4. Drink milk
5. Eat green-leaf food

how to calculate platelets?

Platelets calculate by the following formula.
Platelet counting Formula: N* 20 * 10 9

why it is called improved neubauer chamber?

The Improved Neubauer Chamber is named after its creator, Hans Neubauer, a German physician and inventor. It is a device used in hematology to count cells and particles in a microscopic field.

what is a normal platelet count in newborn?

Generally, a platelet count between 150,000 and 450,000 platelets per microliter of blood is considered within the normal range for newborns.

what is plt in blood test?

PLT in a blood test refers to platelets, which are small blood cells involved in clotting. Platelet count is measured in a complete blood count (CBC) and helps assess clotting ability.

how to write platelet count?

It is typically expressed as a numerical value followed by the unit of measurement, which is “per microliter of blood” (µL). For example 250,000 µL.
